How much spore solution to use? ›

We recommend to use about 10 - 20 ml – that is 1 - 2 syringes - for small bags (2,5 L content). For larger bags (4,5 L content) we recommend to use 20 - 30 ml spore solution – that is 2 - 3 spore syringes. ATTENTION: Should the needle get in contact with any unsterile material, you will have to heat-sterilize it.

Under a flow hood or in a sterile environment, you can inoculate the culture medium with spore solution from your spore syringe. Within a week or two, the spores will germinate and mycelium will begin to colonize the liquid.

One syringe has 20 ml of the mushroom spore solution which is enough to inoculate between 6 and 7 liters of substrate. Drop a few milliliters in each corner of the substrate and everything is set. After some days, the growth of white patches of mycelium can be noticed on the spot where the mushroom spores were dropped.

Colonization occurs after inoculation. This is when the mycelium will start to take over the grain that was inoculated with spores. This period usually lasts between 3 to 6 weeks depending on strain and environmental conditions. Ideal conditions for this phase are in a dark place and temperature between 72 F – 80 F.

Calculate the spore count using the equation: spores/ml = (n) x 10^4, where: n = the average cell count per square of the four corner squares counted .

A: This will vary depending on if you are using spores or a live culture, and also how potent your syringes are. But generally 1-3 ccs of solution is sufficient to inoculate each 1 pound bags.

Liquid culture provides a sterile and controlled environment for the mycelium to grow, which allows for faster and more consistent growth compared to spore syringes.

While the above process is a tried and tested way to create liquid cultures, those looking to forgo this method can find a much simpler way using pre-made spore syringes and liquid culture vials. All you need to do is inject around 1ml of spores into the liquid culture vial, then shake.

Indicators of a Bad Spore Syringe

Watch out for cloudy or murky water in your spore syringe; this is a red flag. Good spores should be suspended in clear liquid. If you see things floating that aren't tiny black dots — the actual spores — something's not right. Also, a funky smell is never a good sign.

Growing mycelium should be kept in an ideal temperature range. For example, P. cubensis colonizes most rapidly between 75-80°F (24-27°C). Temperatures higher than this range may kill the mycelium and encourage growth of contaminants, and temperatures lower than this range may slow down colonization.

How deep should the substrate be for a cubensis monotub? ›

You want to have 3-5 inches of substrate. One 3lb injection port bag should readily inoculate one or two moderate sized tubs. Again, this is a great time to make use of your still air box. You want to start by sprinkling about one inch of bulk substrate into your sterilized monotub, or in the liner if you're using one.

The common standard of spawning is 8-10 liters, or 5-6 kilograms of mycelium per ton of phase 2 compost.

Culture syringes are used by more advanced cultivators and mushroom growers. They are a more reliable starting point in growing mushrooms as the mycelium is already established within the syringe and will therefore colonize faster than just spores on their own.

Heat the needle of your spore syringe in a flame and let it cool. Inject around 4ml of spore solution into each jar. You should distribute this over multiple holes, and try to get it down the inside of the jar so it spreads better. If using micropore tape, replace it again once you remove the needle.

Alternating between the spawn and substrate allows for faster colonization than putting all the spawn on top. In supplemented sawdust this typically means shaking the substrate after inoculation to distribute the spawn evenly. The impact of top vs. through-spawning was inadvertently tested at Fungi Ally.

Inoculating grain spawn

Inject approximately 1-2 ml of liquid culture into a quart-sized jar of grain. Use 5-10 ml of liquid culture to inoculate a 5 lb bag of sterilized grain. A jar or bag of grain should be fully colonized in 14-45 days, depending on the mushroom species and environmental conditions.

Making A Spore Syringe In A Sterilized Environment

Reach your hands into the rubber gloves and scrape the spore print into the sterilized, distilled water. Mix it up a bit with a sterilized instrument or use the syringe to mix up the water by pulling a small amount in and pushing it back out repeatedly.

Inoculating Grain from a Syringe

Then you can quickly lift the lid of the grain jar and inject the syringe. The first bit of liquid will cool down the syringe. Usually about 1 – 2 CC's of liquid culture is sufficient per quart sized grain jar.

Put on your gloves and wipe them with rubbing alcohol. Carefully open the packaging that your liquid culture syringe came in, remove the cap on the end of the syringe and attach the needle. Make sure you shake the liquid culture syringe to disperse the mycelium. Wipe the injection point on your substrate with alcohol.
